Katherine MacDonald

Research Topic and Description: Regulatory T cells are a class of T cells which control immune homeostasis. These cells have a fundamental role in regulating immunity to both self and foreign proteins, and hold promise as a cellular therapy to replace standard immune-suppression in the context of organ transplantation. The efficient production of large numbers of Treg cells in culture is currently a key obstacle to the widespread use of Treg-based therapies. My research aims to optimize the expansion of Treg cell cultures through investigating the timing and levels of environmental stimuli and metabolites that most improve Treg production.

Research Topic and Description: Intra-Operative Ultrasound in Orthopedic Surgery. Orthopedic imaging has traditionally relied on ionizing radiation based modalities such as x-ray, fluoroscopy and CT. We are investigating the employment of 3D US as an alternative safer imaging modality for a prospective minimally invasive computer assisted surgery system designed specifically for pre-operative bone fracture assessment and intra-operative guidance in fracture reduction procedures.

Research Topic and Description: Parkinson’s disease (PD) is a major neurodegenerative disease second to Alzheimer’s. While pharmacological treatments are effective for some features of PD, they tend to be ineffective for tremor. Invasive brain surgery is an effective treatment for many PD features, including tremor; however, not all subjects are suitable candidates for surgery. My research focuses on a non-invasive and non-pharmacological approach to treat PD tremor, based on non-invasive, undetectable, externally applied electrical stimulation.

Research Topic and Description: Fluoroscopic Navigation -- Fluoroscopy C-arms are increasingly used intraoperatively to allow minimal invasion during orthopaedic trauma surgeries. Fluoroscopy therefore aids the surgeons in navigating the tools and implants. Radiology technologists who operate the C-arm fluoroscope often need to take "scouting" X-rays before they are able to achieve the desired view as required by the surgeon. This exposes the patients and the operating room staff to additional ionizing radiation. The aim of my project is therefore to develop a virtual fluoroscopy system that will generate artificial X-rays.The system reconstructs 2D radiographs from a 3D preoperative CT scan based on the tracked position of the C-arm relative to the patient.

Internship Projects: RO Risk Analysis and IVAD Incident Investigation
Project Description 1: Risk assessment of all the RO systems used for dialysis in the lower mainland. RO systems are used to provide purified water that is required in the dialysis. More specifically, the disinfection procedure for the RO is studied and the objective is to provide recommendation for improvements in these procedures.
Project Description 2: Intravenous Access Devices (IVAD) are medication reservoirs that are placed inside the chest area of a patient. Medications are injected inside the reservoir to be spread out throughout the patient’s circulatory system. The project started because there was an incident in which a patient required an immediate removal of the IVAD. After removal, surgeons found that the catheter that was attached to the IVAD was leaking. This IVAD was brought to the biomedical department at VGH for medical device investigation.

Research Topic and Description: Multiple Sclerosis (MS) is a chronic, inflammatory and demyelinating disease. Magnetic resonance (MR) imaging is used to assess the progression of the disease and to evaluate the effect of drug therapy. My research involves appreciating MS White matter (WM) and Grey Matter (GM) lesions in conventional brain imaging and meliorating their consequence on non-rigid registration that may lead to brain tissue misclassification.

Spinal cord injuries can be devastating and debilitating, and often occur as a result of head-on impacts during sporting events. The Pro-Neck-Tor features an internal mechanism which directs the neck into a slightly flexed or extended posture during head-first impacts. In doing so, the direct loads to the cervical spine are dissipated and injury can be prevented.

Research Topic and Description: The effect of wedge and slope angle in high tibial osteotomy. People with varus malalignment often develop osteoarthritis in the medial compartment of the tibiofemoral joint. High tibial osteotomy is a procedure intended to unload the medial compartment of the joint, however it is not well understood how the wedge and slope angle of the osteotomy affect the kinematics and dynamics of the knee. This study will examine how various wedge and slope combinations affect the kinematics and contact pressure of the tibiofemoral joint.

Research Topic and Description:The central nervous system (CNS) has to combine many degrees of freedom available in the human musculoskeletal system to generate a coordinated motor behaviour. There is a growing body of work that supports the existence of a synergistic motor control mechanism utilized by the CNS to facilitate motor coordination. The theory of motor synergies suggests that the CNS controls groups of muscles and/or joints rather than controlling each muscle or joint separately, effectively reducing the high dimensionality of motor planning and execution. Recent studies with stroke survivors suggest that motor impairment after stroke is due to the disruption caused by the cortical lesion in the recruitment and the combination of the motor synergies. The objective of my work is to investigate human upper body motor coordination and to demonstrate the viability of synergistic motor control theory in describing the natural upper body movements, as well as quantifying the effects of stroke on motion generation. As part of this proposed work, I will also investigate the process of motor recovery after stroke by describing how motor synergies change as stroke-survivors complete a conventional physical therapy regimen.

Research Topic and Description: Microfluidic Biomechanical Assay for Red Blood Cells Parasitized by Plasmodium falciparum. Malaria, a disease transmitted by mosquito bite, is caused by parasites that spend part of their life in red blood cells (RBCs). The most common and deadly form of this parasite is Plasmodium falciparum. Once this parasite invades a RBC, the cell becomes stiffer. Marie-Eve’s research involves developing a microfluidic device to measure RBC stiffness using microscale funnel-shaped constrictions.

Research Topic and Description: White blood cells, the primary contaminant cell type, are typically smaller and softer than cancer cells. This property enables to isolate cancer cells based on size and deformability without fixation or chemical modification. Sunyoung’s research involves separating rare circulating tumor cells from the blood cells by employing a microfluidic structural ratchet mechanism and using oscillatory flow through an array of funnel-shaped structures.
